Automated GPCR Signaling Studies in 1536-well Format Using the Non-lytic, Live-cell GloSensor cAMP Assay Technology. A Joint Workshop by Promega, Labcyte and BMG Labtech G protein-coupled receptors (GPCRs) play an important role in a variety of biological processes. They are popular targets in drug discovery research, and a well-designed assay can help expedite the discovery of novel candidates. Many pharmaceutical high-throughput screening (HTS) programs desire a simple, sensitive, automatable GPCR-specific, cell-based assay with a wide dynamic range and low susceptibility to interference. In this workshop, we present the new Promega GloSensor™ cAMP Assay, an HTS-friendly bioluminescent assay capable of measuring intracellular cAMP levels in living cells in real-time. The assay is complemented by ultra low volume transfers of compounds into 1536-well assay plates using the Labcyte® Echo® 555 acoustic liquid handler, while GloSensor™ cAMP cells are dispensed using the Labcyte Deerac™ HTS reagent dispenser. Once the cells and compounds are dispensed, the GloSensor™ cAMP Assay is measured using BMG LABTECH’s PHERAstar Plus HTS multi-detection microplate reader. Implementing a miniaturized GPCR assay as demonstrated in this workshop allows cost-effective screening for modulators of GPCR function, a useful tool for HTS labs in the pharmaceutical industry.
